Premarin Tablets (Conjugated Estrogens) - Product information and Uses

Premarin is a medication that contains a mixture of estrogen hormones mainly consisting of estrone (>50%), equilin (15-25%) and equilenin.
Premarin Tablets are used for the treatment of various indications. It is used for the relief of menopausal and postmenopausal symptoms occurring in naturally or surgically induced estrogen deficiency states. Conjugated Estrogens are also used for prevention of osteoporosis in naturally occurring or surgically induced estrogendeficiency states, hypoestrogenism due to hypogonadism, castration, or primary ovarian failure, atrophic vaginitis and vulvar atrophy

This medication is available as Premarin Tablets manufactured by Wyeth, a division of Pfizer and is packaged in 0.3 mg, 0.625 mg, and 1.25 mg pills.

Side Effects of Premarin Tablets (Conjugated Estrogens)

The common side effects of Premarin Tablets include:

  • Headache
  • Breast pain
  • Irregular vaginal bleeding or spotting
  • Stomach or abdominal cramps, bloating
  • Nausea and vomiting
  • Hair loss
  • Fluid retention
  • Vaginal yeast infection

Serious, but less common side effects include:

  • Heart attack
  • Stroke
  • Blood clots
  • Dementia
  • Breast cancer, Cancer of the lining of the uterus (womb), Cancer of the ovary
  • High blood pressure
  • High blood sugar
  • Gallbladder disease
  • Liver problems
  • Enlargement of benign tumors of the uterus ("fibroids")
  • Severe allergic reactions

Premarin Tablets - Contraindications

Premarin® (conjugated estrogens tablets) should not be prescribed to patients with any of the following health conditions:

  • Patients who are hypersensitive to Conjugated Estrogens or to any other ingredients present in the formulation or component of this medicine.
  • Liver dysfunction or disease as long as liver function tests have failed to return to normal.
  • Known or suspected estrogen-dependent malignant neoplasia
  • Endometrial hyperplasia.
  • Known, suspected, or past history of breast cancer.
  • Undiagnosed abnormal genital bleeding.
  • Known or suspected pregnancy
  • Active or past history of confirmed venous thromboembolism or active thrombophlebitis.
  • Active or past history of arterial thromboembolic disease (e.g. stroke, myocardial infarction, coronary heart disease).
  • Partial or complete loss of vision due to ophthalmic vascular disease.
  • Migraine with aura

 

Premarin Tablets - Dosage

The dosage of Premarin Tablets varies as per the symptoms being treated.
Menopausal symptoms:
The recommended dosage is 0.625 mg to 1.25 mg daily, cyclically or continuously as is medically required. The dosage has to be adjusted upward or downward according to severity of symptoms and response of the patient.
Osteoporosis (loss of bone mass):
The recommended dosage is 0.625 mg daily.
Hypoestrogenism due to Female hypogonadism:
0.3 mg to 0.625 mg daily, administered cyclically (e.g., 3 weeks on and 1 week off) or continuously as required. Doses are adjusted depending on the severity of symptoms and responsiveness of the endometrium.
Hypoestrogenism due to Female castration or primary ovarian failure:
1.25 mg daily, cyclically or continuously as required. Adjust dosage upward or downward according to severity of symptoms and response of the patient. For maintenance, adjust dosage to lowest level that will provide effective control.
Atrophic Vaginitis:
0.3 mg to 1.25 mg daily depending upon the tissue response of the individual patient. Administer cyclically or continuously as required.
Vulvar Atrophy
0.3 mg to 1.25 mg daily depending upon the tissue response of the individual patient. Administer cyclically or continuously as required.

Overdosage and Overdose Symptoms of Premarin Tablets (Conjugated Estrogens)

Overdosage with estrogen (Premarin Tablets) may cause nausea, breast discomfort, fluid retention, bloating or vaginal bleeding in women.
There is no specific antidote to treat estrogen overdose and further treatment if necessary should be symptomatic.
